1.0 OBJECTIVE:
To lay down a procedure for determination
of microbial colonies (cfu) from a petri plates during water and environmental
monitoring by using colony counter.
2.0 SCOPE:
This SOP shall be applicable for
counting of colonies (cfu) to digital colony counter installed in microbiology
area of Q.C Lab.
3.0 RESPONSIBILITY:
3.1
Doing: Q.C Analyst/ Microbiologist.
3.2
Checking: Q.C-Manager.
4.0 ACCOUNTABILITY:
4.1
Head of the Q.C Department.
5.0 MATERIAL AND EQUIPMENT:
5.1
Digital colony counter.
6.0 PROCEDURE:
6.1
Make sure that the instrument is clean.
6.2
Operate the apparatus according to SOP.
6.3
Put petri dish one by one on the wolffhuegel grid, adjust
the magnifying glass, and switch on illumination.
6.4
Ensure that the
apparatus digital display is reset.
6.5
Select the square and count one by one each colony by
marking with auto marker pen.
6.6
Record the total number of cfu count from display in
annexure-1.
6.7
Similarly, perform test on all petri dishes. And note down
colonies in annexure-1.
6.8
Switched off after use and clean it,
