1.0 OBJECTIVE:
To
lay down the calibration procedure of Mechanical pipettor for reliable and accurate results during mixing.
2.0 SCOPE:
This
shall be applicable to the Mechanical
pipettor of Quality Control Instrument Laboratory.
3.0 RESPONSIBILITY:
3.1
Technical
Assistant/ Helper.
3.2
Q.C Analyst.
4.0 ACCOUNTABILITY:
4.1
Manager-Quality
Control.
5.0 MATERIAL AND
EQUIPMENT:
5.1
Mechanical pipettor.
6.0 PROCEDURE:
6.1 Maintain
the Mechanical pipettor
according to the SOP
6.2 Operate the Mechanical pipettor according to
the SOP
6.3 Calibration
should be done every 3 month with weight variation.
6.4 Different
set of volumes should be taken and linearity should be performed.
6.5 Attach
the micropipette tips to micropipette. The desired volume is set and water
shall be aspirated. Dispensed into dry beaker and weighed repeat for 10 times.
The mean of the weights are converted into μl using the following formula:
Volume
= _____Weight of water sample._____
Density of water (at 25oC)
6.6 The percentage variation can be calculated by
using following formula:
Percentage
variation = Actual weight – Observed weight x 100
Actual
weight
6.7 Perform the calibration by suing the set
volume.
6.8 Record
the result on the “Calibration Record”,
“Calibration Log Book” & paste the “Calibration
Status Sticker” on Mechanical
pipettor.
6.9 Precautions:
6.9.1 Use
the analytical balance with a readability of 0.1mg.
