Automatic Polarimeter Operational SOP



1.0  OBJECTIVE:
To lay down the operating procedure of Polarimeter.
2.0  SCOPE:
This shall be applicable to the Polarimeter of Quality Control Laboratory.
3.0  RESPONSIBILITY:
3.1  Technical Assistant/ Helper.
3.2  Q.C Analyst.
4.0  ACCOUNTABILITY:
4.1  Manager-Quality Control.
5.0  MATERIAL AND EQUIPMENT:
5.1  Polarimeter.
5.2  Sample.
5.3  Solvent as specified in monograph (as blank).
6.0  PROCEDURE:
6.1  Ensure that the instrument is clean. Follow the SOP.
6.2  Switch ON the mains.
6.3  Switch ON the Polarimeter.
6.4  Wait till the sodium lamp glass with full intensity of yellow light.
6.5  Adjust vernier scale to zero dial scale to zero.
6.6  View through the piece, if there is difference in intensity of light adjust it without disturbing the dial reading by adjusting, the knurled screw of the polarize prism.
6.7  It is different to adjust with knurled screw, adjust the dividing line by rating dial and vernier scale. Note down the reading, which is dextro and levo and will be taken as zero error for the calculation of the optical rotation.
6.8  Rinse the Polarimeter tube with blank solution.
6.9  Fill the tube with blank solution at specified temperature.
6.10    Clean the sides of the tube with tissue paper.
6.11    If any air bubble comes then it must be trapped in center of Polarimeter tube.
6.12    Keep the Polarimeter tube in its place in the Polarimeter.
6.13    Adjust the zero on the upper scale with the help of control wheel.
6.14    Focus circular disc of the light with the help of field telescope.
6.15    By means of control wheel, move the scale from its first position.
6.16    Observe the field, which is divided into two valves, light and dark.
6.17    Adjust the control wheel to get equal intensity of the two halves.
6.18    If the index line between two scales division, therefore rotate the knurled wheel to bring the index line coincident with a scale line.
6.19    Note the reading in degree of rotation.
6.20    Repeat the operation 5 times.
6.21    Consider the average reading.
6.22    Mark it as + (plus) reading if it is left to the zero and – (minus) reading if it is right to the zero.
6.23    Remove the Polarimeter tube from the Polarimeter.
6.24    Discard the solution from the Polarimeter tube.
6.25    Rinse the Polarimeter tube twice with the test solution.
6.26    Fill the test solution in the Polarimeter tube at the temperature specified in the monograph/ specification.
6.27    Clean the tube and the sides of glass with tissue paper.
6.28    If any air bubbles come then it must be trapped in the centre of Polarimeter tube.
6.29    Keep the tube in its place in the Polarimeter.
6.30    Adjust the control wheel to obtain same field condition as with the blank and note down the reading.
6.31    Repeat the operation at least 5 times.
6.32    Take the average of the reading.
6.33    Discard the test solution from the Polarimeter tube.
6.34    Switch OFF the mains.
6.35    Clean the Polarimeter tube with distilled water if solutions are prepared in polar solution or clean the Polarimeter tube with methanol or ether if solvent is non-polar.
6.36    Keep the tube in tube box.
6.37    Enter the details in logbook.
6.38    Precaution:
6.38.1    Polarimeter tube should be filled in such a way so as to avoid air bubbles.
6.38.2    Before taking the reading clean the Polarimeter tube with tissue appear.
6.38.3    Set the field telescope for better observation.
6.38.4    The optical elements must be cleaned and at exact alignment.
6.38.5    Tighten the end plate with caps and seal so as to make it leak proof.
